Generic Name: sitagliptin and metformin HCl 

Drug Class: dipeptidyl peptidase-4 (DPP-4) inhibitor and biguanide 


Janumet XR is an oral prescription drug used to improve glycemic control among adults with type 2 diabetes mellitus when treatment with sitagliptin and metformin is necessary. This is prescribed together with proper diet and exercise.  

Janumet XR is not indicated for type 1 diabetes and diabetic ketoacidosis. It has not been investigated in patients who have had pancreatitis in the past. 

Dosage Forms 

This is available in Janumet XR 50 1000 mg, 50/500 mg, and 100/1000 mg extended-release tablets. 

Janumet XR Mechanism of Action 

This is a combination product of a DPP-4 inhibitor and a biguanide. It slows the inactivation of incretin hormones for starters. By prolonging and increasing the levels of active incretin hormones, insulin release is increased, and glucagon levels in the circulation are decreased in a glucose-dependent manner. Secondly, it reduces the production of hepatic glucose, reduces gastrointestinal glucose absorption, and improves insulin sensitivity through increased peripheral glucose uptake and utilization. 

How to Take Janumet XR 

Before starting to use Janumet XR, read the information leaflet or medication guide provided by your healthcare professional. The only way to take this tablet is by mouth. This is usually prescribed once a day, preferably in the evening. You must take it with a meal. Swallow the extended-release tablet entirely and drink plenty of fluids. Cutting or crushing this product is not recommended.
